Notch and Wnt regulate intestinal stem cells differentiation balance

  Recently, the international academic journals of cell reports published a latest research progress of American scientists, they found that in small intestine stem cells and intestinal homeostasis regulation process, Notch and Wnt signaling played an important regulatory role , blocking the Notch signaling would trigger intestinal stem cells differentiation imbalance, and accompanied by disinhibition of the Wnt signaling pathway in the process.Organ homeostasis balance should strictly adjust adult stem cells , and by a variety of signals integration to realize the differentiation process of stem cells into terminal cell .

    In the small intestine of mice, Notch and wnt signaling are very necessary for intestinal stem cell maintenance and differentiation balance between intestine cells responsible for digestive juice secretion and cells responsible for nutrients absorption . The researchers found that when Notch signaling was inhibited , intestinal stem cells tend to differentiate into secretion cells, and thus affecting differentiation process of nutrients absorption cells . The researchers used antibody with functional blocking properties specifically blocked Notch receptor and they found that blocking the Notch signaling would relieve the inhibition of the Wnt signaling pathway, disrupting the normal function of intestinal stem cells, leading to erroneous expression of pro-secretory function related genes .

   Meanwhile, the researchers also found inhibition of the wnt signaling pathway could restore Notch signaling blockade resulting phenotype. This study clarified a negative adjustment mechanism that maintain the activity of stem cells and differentiation balance , and it also proved that the interactions between the Wnt and Notch signaling was very important to maintain normal function in adult stem cells .
